In emails or letters it's short form of reply, but is it also used for (RE) regarding ...... some matter in some official letter heads?

According to the Oxford Shorter dictionary, "Re" is short for regarding. This is not a learner's dictionary.

It means: about the subject mentioned or regarding the subject mentioned.
