Nonspecific haze or film over entire antigen substrate in all controls and specimens.
POSSIBLE CAUSES
Antigen substrate inadequately rinsed and washed between incubation.
Drying of antigen substrate after rinse and wash steps.
Check fluorescence microscope. Optical pathway may be dirty.
Check if coverslip is too thick or if two coverslips are stuck together.
POSSIBLE SOLUTIONS
Frequent changes of fresh PBS for the rinse and wash steps are necessary.
Do not allow antigen to dry at any time during or between staining steps. Handle only one or two slides at a time from wash step before applying conjugate and returning to moist chamber.
Clean objectives and eyepiece lens.
Use #1 coverslip. Carefully remove double coverslip and remount with single coverslip.
Translation - English PROBLEMA
Presenza di torbidità (haze) o patina aspecifiche nell’intero substrato antigenico di ogni campione e di tutti i controlli.
POSSIBILI CAUSE
Il substrato antigenico non è stato risciacquato e lavato in maniera adeguata tra le varie fasi di incubazione.
Il substrato antigenico si è asciugato dopo le fasi di risciacquo e lavaggio.
Controllare il microscopio a fluorescenza per assicurarsi che il percorso ottico non sia sporco.
Controllare il vetrino coprioggetti per assicurarsi che non sia troppo spesso o che non si siano incollati due vetrini.
POSSIBILI SOLUZIONI
Per espletare le fasi di risciacquo e lavaggio è necessario cambiare di frequente il tampone fosfato salino (PBS).
Non permettere mai all’antigene di asciugarsi durante le fasi di colorazione, o tra una fase e l’altra. Manipolare soltanto uno o due vetrini alla volta, dalla fase di lavaggio, all’applicazione del coniugato, alla riposizione in camera umida.
Pulire gli obiettivi e l’oculare. Utilizzare 1 vetrino coprioggetti. Rimuovere attentamente il doppio vetrino e rimontare con un singolo vetrino.
