successive cycles Explanation: After amplification at 94oC for 5 min, followed by successive cycles at 94oC for 30 sec, 60oC for 30 sec, and 72oC for 30 sec for 30 cycles, followed by a final extension reaction at 72oC for 7 min, samples of purified products from reactions #1 and 2 were combined as templates in PCR reaction #3, using T7 promoter and terminator primers as amplimers. Corresponding proCrp4 templates of these variants were prepared as described (16).
